Staining is used to highlight the important features of the tissue and to enhance the tissue contrast in histology. Hematoxylin and Eosin are the most widely used dyes in tissue staining which stains the cell’s nucleus to bluish and pinkish colours. The first staining step is de-waxing which uses solvent to remove the wax from the slide before staining When a tissue is completely stained, it is covered with cover-glass which makes preparation permanent.
